The five-component acellular pertussis vaccine is a protein subunit vaccine developed by Connaught Laboratories (now Sanofi Pasteur) for the prevention of pertussis (whooping cough) caused by *Bordetella pertussis*. It contains five highly purified antigenic components: glutaraldehyde-inactivated pertussis toxin (pertussis toxoid), filamentous hemagglutinin (FHA), pertactin (PRN), and fimbriae types 2 and 3 (FIM2/3). This vaccine was developed as a safer alternative to whole-cell pertussis vaccines, demonstrating approximately 85.2% efficacy in clinical trials with a significantly lower incidence of adverse events such as fever and local reactions. It is typically administered as the pertussis component of combination vaccines including DTaP (e.g., Daptacel) and Tdap (e.g., Adacel), often adsorbed to an aluminum-based adjuvant to enhance the immune response.
