FK-960 is a small molecule drug originally developed as a potential cognitive enhancer and anti-dementia agent. It acts primarily as an agonist at somatostatin receptors (SSTR) and also exhibits agonist activity at serotonin (5-hydroxytryptamine or 5‑HT) receptors. Preclinical studies indicate that it stimulates both somatostatin and serotonin production in the brain and enhances the release of glial cell line-derived neurotrophic factor (GDNF) via ERK-dependent pathways in astrocytes. These actions are believed to underlie its proposed cognitive-enhancing effects. The compound was investigated for use in Alzheimer's disease, dementia-related disorders including schizoaffective disorder and schizophrenia-associated cognitive impairment but development was discontinued after Phase 2 clinical trials[1][3][5][6][9]. Chemically it is a benzamide derivative with a fluorine atom at the para position of the phenyl ring and an acetyl-piperazinyl group attached to the amide nitrogen[10]. The original developer was Fujisawa Pharmaceutical (now part of Astellas Pharma). No approved indications; all development has been discontinued.
