Overview

FK1706 is a novel, non-immunosuppressive immunophilin ligand derived from FK506 (tacrolimus). Unlike its parent compound, it does not suppress the immune system but exhibits significant neurotrophic and neuroregenerative activity. Its mechanism of action involves binding to the immunophilin protein FKBP-52 and modulating the RAS/RAF/MAPK signaling pathway. Preclinical studies have demonstrated that FK1706 can ameliorate painful diabetic neuropathy by restoring intraepidermal nerve fiber density and improving mechanical allodynia without affecting blood glucose levels. It has also shown efficacy in enhancing nerve regeneration and functional recovery in models of nerve injury, including bladder function recovery after neurorrhaphy and erectile function following cavernous nerve injury[1][2][5][6][9]. Clinical trials have been conducted to assess its effects on epidermal nerve fiber regeneration in humans[8].
