FK409 is a small molecule nitric oxide donor originally developed as a novel vasodilator. It releases nitric oxide (NO) spontaneously, with a half-life of approximately 40 minutes at physiological pH and temperature. FK409 acts as a potent vasodilator, leading to increased cyclic guanosine monophosphate (cGMP) in vascular smooth muscle, and has shown pharmacological effects similar to organic nitrates like nitroglycerin, including dilation of coronary arteries. It was developed for the treatment of angina pectoris, but clinical development was terminated primarily due to the occurrence of drug-induced anemia. The compound has also been studied for its ability to enhance morphine analgesia peripherally and reduce portal hypertension and liver injury in small-for-size liver transplantation models[3][4][1][6][7].
