Overview
FL-302 is a preclinical-stage tetravalent bispecific antibody designed to target Claudin 18.2 (CLDN18.2) and CD137 (4-1BB). Originally developed by NovaRock Biotherapeutics and licensed by Leap Therapeutics through its acquisition of Flame Biosciences, the molecule is engineered to provide conditional activation of the CD137 immune-stimulatory pathway specifically in the presence of CLDN18.2-expressing tumor cells. This localized agonism is intended to enhance T-cell mediated anti-tumor activity within the tumor microenvironment while minimizing the systemic toxicities typically associated with broad CD137 activation. FL-302 is being developed for the treatment of advanced solid tumors, with a particular focus on gastric and pancreatic cancers where CLDN18.2 is frequently overexpressed.
