FL-BAI3 (Full-length Brain-specific Angiogenesis Inhibitor 3) is a protein isoform of the adhesion G-protein-coupled receptor BAI3, encoded by the *ADGRB3* gene. Predominantly expressed in the brain, BAI3 is involved in regulating dendritic morphogenesis and synaptic plasticity. In the context of glioblastoma multiforme (GBM), FL-BAI3 expression is significantly downregulated due to epigenetic hypermethylation of its regulatory regions. Research indicates that FL-BAI3 acts as a tumor suppressor by inhibiting the epithelial-to-mesenchymal transition (EMT) signature and attenuating tumor growth in glioblastoma cell lines and orthotopic mouse models. However, preclinical survival analysis suggests that while FL-BAI3 attenuates tumor growth, it may not confer the same survival benefit as the shorter S-BAI3 isoform. It is currently being investigated for its potential in isoform-specific epigenetic reactivation and therapeutic targeting in GBM.
