Overview

FL1 FAPα CAR-T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target Fibroblast Activation Protein alpha (FAPα), a protein highly expressed on cancer-associated fibroblasts (CAFs) and certain tumor cells within the solid tumor microenvironment. Developed by Eutilex using the proprietary Eumbody System (a high-throughput scFv library), the FL1 clone features an optimized binding affinity intended to overcome common CAR-T limitations such as trogocytosis and T-cell exhaustion. By utilizing a CD28/CD3ζ signaling architecture and a lower structural avidity scFv, FL1 CAR-T cells aim to preserve target antigen expression and maintain a more persistent, less exhausted phenotype. Preclinical evaluations in glioblastoma models have demonstrated the potential of these cells to penetrate the tumor microenvironment and exert anti-tumor effects.
