FL115 is an engineered, long-acting interleukin-15 (IL-15) superagonist developed by Suzhou Forlong Biotechnology. It is a recombinant fusion protein consisting of a mutated IL-15 (N72D) and the sushi domain of the IL-15 receptor alpha (IL-15Rα) fused to an Fc region. This design mimics the trans-presentation of IL-15, significantly enhancing its potency and half-life compared to recombinant human IL-15. FL115 selectively activates the IL-15 receptor complex on natural killer (NK) cells and CD8+ effector T cells, promoting their proliferation and anti-tumor activity while minimizing the stimulation of regulatory T cells (Tregs). It is currently in clinical development for advanced solid tumors as both a monotherapy and in combination with checkpoint inhibitors, as well as for non-muscle invasive bladder cancer.
