FL496 is a small molecule antineoplastic agent developed by Canget BioTekpharma. It is a derivative of FL118, a camptothecin analog known for its ability to inhibit multiple anti-apoptotic proteins. FL496 specifically targets the DDX5 (p68) pathway, a DEAD-box RNA helicase that is frequently overexpressed in various malignancies and plays a critical role in tumor cell survival and proliferation. By modulating this pathway, FL496 facilitates the downregulation of key survival factors such as survivin, Mcl-1, XIAP, and cIAP2. Preclinical studies have demonstrated its efficacy in in vivo models of retinoblastoma and in vitro models of type 2 kidney cancer and malignant pleural mesothelioma.
