FL8 FAPα CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target fibroblast activation protein alpha (FAPα), a marker predominantly expressed on cancer-associated fibroblasts (CAFs) in the tumor microenvironment and on certain solid tumor cells. Developed by Optieum Biotechnologies using the proprietary Eumbody System, FL8 is a second-generation CAR-T (utilizing CD28 and CD3ζ signaling domains) that features an optimized scFv with reduced structural avidity compared to its parental clone. This affinity tuning is intended to mitigate trogocytosis—the process where T cells strip antigens from target cells—thereby reducing T-cell exhaustion and enhancing the persistence and efficacy of the therapy in treating refractory solid malignancies such as glioblastoma.
