FL8 is an engineered chimeric antigen receptor (CAR) T-cell therapy designed to target Fibroblast Activation Protein alpha (FAPα), a protein highly expressed on cancer-associated fibroblasts (CAFs) and certain solid tumor cells. Developed by Eumunity using the proprietary Eumbody System, FL8 features an optimized single-chain variable fragment (scFv) with tuned binding affinity. This reduced structural avidity is intended to mitigate trogocytosis—the unintended stripping of antigen from target cells—and reduce T-cell exhaustion, thereby improving the persistence and efficacy of the treatment within the immunosuppressive tumor microenvironment. FL8 is being investigated for the treatment of refractory solid malignancies, including glioblastoma, where it aims to concurrently target tumor cells and the supportive CAF-rich microenvironment.
