Overview

Flagrp170 is a next-generation, multifunctional chimeric chaperone-based immunolytic agent developed for cancer immunotherapy. It is a recombinant fusion protein consisting of the large heat shock protein/chaperone grp170 (glucose-regulated protein 170) and a flagellin-derived NF-κB-stimulating sequence. The agent is designed to enhance the cross-presentation of tumor-associated antigens and provide potent co-stimulation to the immune system via Toll-like receptor 5 (TLR5) activation. Administered intralesionally, Flagrp170 reprograms the tumor microenvironment (TME) by inducing a Th1-polarized phenotype characterized by the elevation of cytokines such as GM-CSF, IL-12, and IFN-γ. This process facilitates the recruitment and activation of dendritic cells and CD8+ T cells, leading to systemic antitumor immunity. Preclinical studies have demonstrated its efficacy in various mouse models, including melanoma, prostate cancer, colorectal cancer, and mammary tumors.
