Flanvotumab (IMC-20D7S) is a recombinant human IgG1 monoclonal antibody that targets tyrosinase-related protein 1 (TYRP1, also known as gp75), a transmembrane glycoprotein specifically expressed in melanocytes and melanoma cells. By binding to TYRP1 on the surface of melanoma cells, flanvotumab induces immune-mediated antitumor effects primarily through antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). These mechanisms recruit natural killer cells and other immune effectors to destroy TYRP1-positive tumor cells. Flanvotumab was developed by ImClone Systems and Eli Lilly for the treatment of advanced melanoma. In phase I clinical trials, it demonstrated a favorable safety profile with evidence of antitumor activity in patients with relapsed or refractory melanoma[2][4][5][8].
