Overview

Fletikumab (NNC0109-0012) is a monoclonal antibody that targets interleukin-20 (IL-20), a pro-inflammatory cytokine involved in the pathogenesis of several autoimmune and inflammatory conditions. Originally developed by ZymoGenetics and later acquired by Novo Nordisk, fletikumab was investigated as a biologic therapy for moderate to severe chronic plaque psoriasis, rheumatoid arthritis, and psoriatic arthritis. By binding to and neutralizing IL-20, the drug aims to inhibit the signaling pathways that drive keratinocyte hyperproliferation and synovial inflammation. Although Phase 1 clinical trials demonstrated that fletikumab was well-tolerated with a pharmacokinetic profile typical of monoclonal antibodies, its clinical development for psoriasis and rheumatoid arthritis has been discontinued.
