Overview

FLLL12 is a synthetic curcumin analog developed to enhance the potency and pharmacokinetic properties of curcumin. It functions primarily as an activator of the NRF2 (Nuclear factor erythroid 2-related factor 2) signaling pathway, which orchestrates the cellular antioxidant response. By promoting the expression of cytoprotective genes such as HMOX-1, NQO1, and SLC7A11, FLLL12 helps mitigate oxidative stress and protect cells from carcinogenic damage. Research has demonstrated its efficacy in normal, premalignant, and malignant head and neck cancer cell lines, highlighting its potential as a chemopreventive agent. FLLL12 is reported to have a superior pharmacokinetic profile compared to natural curcumin.
