FLOT + PD-1 antibody is a combination regimen consisting of the chemotherapeutic protocol known as FLOT (docetaxel, oxaliplatin, leucovorin, and 5-fluorouracil) together with a programmed cell death protein 1 (PD-1) monoclonal antibody. The FLOT regimen is established as a preferred perioperative chemotherapy for localized gastric and gastroesophageal junction adenocarcinoma. The addition of a PD-1 inhibitor—such as sintilimab or nivolumab—aims to enhance antitumor immune responses by blocking the inhibitory PD-1 pathway on T cells, thereby promoting immune-mediated tumor cell killing. This combination is under investigation primarily for its potential to improve pathological complete response rates and survival outcomes in patients with advanced or resectable gastric or gastroesophageal junction adenocarcinoma[2][3][4]. Mechanistically, the chemotherapy component induces cytotoxicity against rapidly dividing tumor cells while the anti-PD-1 agent relieves immunosuppression within the tumor microenvironment.
