Drug intelligence / Profile preview
Floxuridine liposome refers to a drug delivery system where floxuridine, an antimetabolite, is encapsulated within liposomes. This formulation aims to improve the drug's pharmacokinetics, stability, and tumor targeting, potentially enhancing efficacy and reducing toxicity compared to free floxuridine. Floxuridine itself is a pyrimidine analog that is rapidly catabolized to 5-fluorouracil (5-FU), which interferes with DNA synthesis by inhibiting thymidylate synthetase and is incorporated into RNA, leading to the production of fraudulent RNA. Prominent examples of floxuridine liposome formulations include combination drugs that also encapsulate irinotecan, a topoisomerase I inhibitor. Irinotecan is a prodrug that is converted to its active metabolite, SN-38, which blocks the topoisomerase I enzyme, resulting in DNA damage and cell death. These liposomal combination products are being developed for various cancers, particularly advanced gastrointestinal cancers and colorectal cancer.
