FLT3 CAR-NK cells are an experimental, allogeneic cell-based immunotherapy designed for the treatment of acute myeloid leukemia (AML). These natural killer (NK) cells are genetically engineered to express a chimeric antigen receptor (CAR) that specifically targets FMS-like tyrosine kinase 3 (FLT3), a protein frequently overexpressed on AML blasts and leukemic stem cells. To enhance therapeutic efficacy, the cells are often modified to secrete soluble interleukin-15 (IL-15), which supports NK cell survival, persistence, and expansion in the tumor microenvironment. Developed as an off-the-shelf product, this therapy aims to provide a readily available alternative to patient-specific CAR-T therapies. Preclinical research conducted at City of Hope and by CytoImmune Therapeutics has demonstrated that these cells can effectively eliminate FLT3-positive leukemia cells while sparing healthy hematopoietic stem cells.
