Overview

FLT3 CAR_sIL-15 NK cells (CYTO-102) is an allogeneic, off-the-shelf natural killer (NK) cell therapy engineered to express a chimeric antigen receptor (CAR) targeting FMS-like tyrosine kinase 3 (FLT3) and to secrete soluble interleukin-15 (sIL-15). Developed by CytoImmune Therapeutics in collaboration with City of Hope, this therapeutic candidate is designed for the treatment of relapsed or refractory acute myeloid leukemia (AML). The integration of sIL-15 is intended to enhance the persistence and metabolic fitness of the NK cells in the immunosuppressive tumor microenvironment and to provide paracrine activation of endogenous T cells and NK cells. The construct incorporates a CD28/CD3ζ costimulatory domain and a truncated EGFR (tEGFR) safety switch, which allows for the rapid depletion of the engineered cells via cetuximab administration if needed. Preclinical studies have demonstrated potent antileukemic activity and improved survival in AML models without significant toxicity to healthy hematopoietic stem cells.
