Flt3 ligand (FLT3L) is a hematopoietic cytokine and growth factor that binds to the fms-like tyrosine kinase 3 (FLT3/CD135) receptor on hematopoietic progenitor cells. This interaction induces dimerization and activation of the receptor's intrinsic tyrosine kinase activity, leading to autophosphorylation and downstream signaling through pathways such as STAT5a, ERK1/2, and PI3K[6][8]. FLT3L acts synergistically with other growth factors to stimulate proliferation and mobilization of bone marrow precursor cells—including CD34+ stem cells—and promotes differentiation into dendritic cells as well as B cell and T cell lineages[1][4][7]. Recombinant forms of FLT3L are used experimentally for ex vivo expansion or mobilization of hematopoietic stem/progenitor cells. The protein is structurally homologous to stem cell factor (SCF) and colony stimulating factor 1 (CSF-1)[7].
