Flt3 ligand-Fc is a recombinant fusion protein consisting of the Fms-like tyrosine kinase 3 (Flt3) ligand joined to the Fc region of an immunoglobulin. This molecule acts as a potent agonist for the Flt3 receptor (CD135), which is primarily expressed on hematopoietic progenitor cells, dendritic cell precursors, and acute myeloid leukemia (AML) blasts. Binding of the ligand to the receptor triggers dimerization and activation of downstream signaling pathways, including PI3K/AKT and MAPK/ERK, which can stimulate cell cycle progression and survival. In therapeutic development, Flt3 ligand-Fc is utilized to expand dendritic cell populations for immunotherapy or as a targeting moiety in ligand-drug conjugates (LDCs), such as FL-Fc-DM1, to selectively deliver cytotoxic payloads like mertansine directly to Flt3-expressing malignant cells, potentially overcoming resistance in quiescent leukemia stem cells.
