Overview

Flubentylosin (ABBV-4083) is a next-generation oral small molecule macrolide antibiotic derived from tylosin A and developed as an anti-Wolbachia agent for the treatment of filarial diseases such as onchocerciasis (river blindness) and lymphatic filariasis. It acts by inhibiting protein synthesis in Wolbachia endosymbionts essential for the survival and fertility of filarial worms. Flubentylosin demonstrates superior efficacy to doxycycline in preclinical models, with improved oral bioavailability and a shorter required dosing regimen. The drug was developed through collaboration between AbbVie and the Drugs for Neglected Diseases Initiative (DNDi), with clinical trials conducted up to Phase II before discontinuation[1][2][6][7].
