Overview

Vocimagene amiretrorepvec/flucytosine is an experimental combination therapy consisting of a gene therapy agent and a prodrug. Vocimagene amiretrorepvec (Toca 511) is a retroviral replicating vector that delivers a gene encoding cytosine deaminase to cancer cells. When followed by administration of flucytosine (Toca FC), the expressed enzyme converts flucytosine into the anticancer agent 5-fluorouracil (5-FU) within the tumor cells. This approach allows for local production of 5-FU in the tumor microenvironment, killing cancer cells and immunosuppressive myeloid cells, which leads to T-cell-mediated antitumor immune activity. The therapy is primarily being developed for high-grade gliomas and other brain cancers.
