A combination chemotherapy regimen used primarily for the treatment of acute myeloid leukemia (AML), particularly in CD33-positive patients. This regimen combines the nucleoside analogs fludarabine and cytarabine with the anthracycline idarubicin and the antibody-drug conjugate gemtuzumab ozogamicin. The combination has shown high overall response rates and good disease debulking capabilities in clinical trials [1]. Studies have shown that it improves event-free survival in certain AML patient populations compared to other treatment approaches [2]. The therapy is particularly effective in patients with specific genetic mutations, including NPM1 and FLT3 mutations [2][4]. For core binding factor (CBF) acute myelogenous leukemia, a variant without idarubicin (FLAG-GO) showed superior results compared to FLAG-IDA [3]. It represents an active treatment strategy for CD33-positive AML patients, particularly those aged 65 years or younger, offering high overall response rates and favorable disease control.
