Fluocinonide is a potent synthetic glucocorticoid corticosteroid used topically as an anti-inflammatory and antipruritic agent for the treatment of various skin disorders such as eczema and psoriasis. It works by binding to the cytosolic glucocorticoid receptor in skin cells; this complex then translocates to the cell nucleus where it modulates gene expression by binding to glucocorticoid response elements (GREs). This leads to increased synthesis of lipocortins (such as annexin-1), which inhibit phospholipase A2 and thereby reduce production of inflammatory mediators like prostaglandins and leukotrienes. The drug also suppresses immune responses locally in the skin. Fluocinonide is classified as a high or super-high potency topical corticosteroid depending on formulation concentration. It was first approved in 1971 for use in adults and children over 12 years old[1][2][4][5].
