Overview

Fluoromethylcarnitine (FMC) is a small molecule inhibitor of the bacterial enzyme trimethylamine (TMA) lyase. It is designed to block the microbial metabolism of dietary carnitine into TMA, which is subsequently converted by host hepatic enzymes into trimethylamine N-oxide (TMAO). Elevated TMAO levels are associated with various inflammatory and cardiovascular pathologies, including atherosclerosis and rheumatoid arthritis. In preclinical research, specifically in murine models of collagen-induced arthritis (CIA), FMC has been demonstrated to effectively reduce intestinal TMA and circulating TMAO levels. While its efficacy in ameliorating arthritis symptoms in some studies has been noted as less potent than other modulators like 3,3-dimethyl-1-butanol (DMB), it remains a significant tool for investigating the gut-microbiome-immune axis. The compound is primarily developed and studied within academic research settings, such as the University of Colorado Anschutz Medical Campus.
