Fluorouracil is a pyrimidine analog and antimetabolite used as an antineoplastic agent in the treatment of various cancers. It works primarily by inhibiting thymidylate synthase after conversion to its active metabolite (FdUMP), thereby blocking DNA synthesis and leading to cell death. Fluorouracil can also be incorporated into RNA in place of uridine triphosphate (UTP), disrupting RNA processing and protein synthesis. It is used systemically for cancers such as colorectal cancer, breast cancer, gastric cancer, pancreatic cancer and topically for actinic keratosis and basal cell carcinoma[1][3][4][6].
