A combination of sustained-release formulations of the antimetabolite 5-fluorouracil (5-FU) and the platinum-based chemotherapeutic cisplatin. This specific formulation is designed for intraoperative implantation into the liver incisal margin following hepatectomy in patients with high-risk hepatocellular carcinoma (HCC). The sustained-release delivery system aims to maintain high local drug concentrations at the surgical site to prevent tumor recurrence while minimizing systemic toxicity. 5-FU acts by inhibiting thymidylate synthase, disrupting DNA and RNA synthesis, while cisplatin forms DNA crosslinks that induce apoptosis. This approach was investigated in clinical trials sponsored by the Eastern Hepatobiliary Surgery Hospital in China, often in conjunction with postoperative transarterial chemoembolization (TACE).
