Overview

It is used primarily in the treatment of advanced or metastatic cancers, including colorectal cancer and gastric cancer, particularly in patients who have progressed after standard therapies[1][3][8]. - **Fluorouracil** is an antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis and repair, as well as interfering with RNA processing. - **Folinic acid** enhances the binding of fluorouracil’s active metabolite to thymidylate synthase, increasing its cytotoxic effect[5]. - **Mitomycin C** is an alkylating agent that crosslinks DNA strands, inhibiting DNA synthesis and function. The combination exploits synergistic effects to increase anti-tumor activity against rapidly dividing cells. This regimen has been studied for use in heavily pretreated metastatic colorectal cancer patients and advanced gastric cancer[1][8].
