Overview
Fluorouracil + irinotecan is a combination of two chemotherapy drugs commonly used in the treatment of various cancers, most notably colorectal (bowel) cancer. Fluorouracil (also known as 5FU) is an antimetabolite that inhibits thymidylate synthase, thereby interfering with DNA synthesis and function. Irinotecan is a topoisomerase I inhibitor that prevents DNA from unwinding and replicating by stabilizing the complex between topoisomerase I and DNA, leading to double-strand breaks and cell death. This combination exploits different mechanisms to target rapidly dividing cancer cells[1][2][5]. The regimen may be administered alone or as part of multi-drug protocols such as FOLFIRI (which also includes folinic acid/leucovorin)[1][2].
