Overview

This is a combination chemotherapy regimen consisting of three agents: - **Fluorouracil (5-FU):** An antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death. - **Leucovorin calcium (folinic acid):** A reduced folate that enhances the binding of fluorouracil to thymidylate synthase, increasing its cytotoxicity. - **Vorinostat (suberoylanilide hydroxamic acid/SAHA):** A histone deacetylase inhibitor that alters gene expression by increasing acetylation of histone proteins, leading to cell cycle arrest and apoptosis in cancer cells. This combination has been investigated primarily for the treatment of metastatic or refractory colorectal cancer. Clinical studies have shown activity in patients with fluorouracil-refractory disease. The regimen is under clinical development and not approved as a fixed combination product; each component is individually approved for use in oncology[1][2][3].
