Fluorouracil + mitomycin c is a combination chemotherapy regimen consisting of two cytotoxic agents—fluorouracil (also known as 5-fluorouracil or 5FU) and mitomycin c. This combination is most commonly used in conjunction with radiotherapy (chemoradiation) for the treatment of nonmetastatic anal canal cancer and muscle-invasive bladder cancer. Fluorouracil is a pyrimidine analog that inhibits thymidylate synthase, thereby interfering with DNA synthesis and function. Mitomycin c acts as an alkylating agent that crosslinks DNA, leading to inhibition of DNA synthesis and function. The synergistic effect of these drugs enhances the cytotoxic impact on rapidly dividing tumor cells when combined with radiation therapy[1][2][4][5]. The FUMIR regimen refers specifically to this drug pair given concurrently with radiation.
