Flutafuranol (also known as NAV4694 or AZD4694) is a next-generation fluorine-18 ([18F]) labeled radiopharmaceutical diagnostic agent developed for positron emission tomography (PET) imaging of cerebral beta-amyloid (Aβ) plaques. Originally discovered by AstraZeneca and later developed by Navidea Biopharmaceuticals and Meilleur Technologies (acquired by Lantheus in 2024), flutafuranol exhibits high affinity and selectivity for Aβ fibrils, a hallmark pathological feature of Alzheimer's disease. Compared to first-generation tracers like [11C]PiB, flutafuranol offers the logistical advantages of the longer half-life of fluorine-18 (approximately 110 minutes) and demonstrates lower non-specific binding in white matter. This improved signal-to-noise ratio allows for more accurate detection and quantification of brain amyloid burden, which is critical for the early diagnosis of Alzheimer's disease, patient stratification in clinical trials, and monitoring the efficacy of disease-modifying anti-amyloid therapies.
