fluticasone + omeprazole

This combination therapy consists of the proton pump inhibitor (PPI) omeprazole and the topical corticosteroid fluticasone. It is being investigated for the treatment of eosinophilic esophagitis (EoE). While omeprazole is traditionally used for acid suppression, in the context of EoE, it has been shown to inhibit STAT6 binding to the eotaxin-3 promoter, thereby reducing the recruitment of eosinophils to the esophageal mucosa through an anti-inflammatory mechanism independent of its effect on gastric acid. Fluticasone acts as a local anti-inflammatory agent to further reduce eosinophilic infiltration. The combination is being evaluated in a Phase 4 clinical trial (NCT03781596) sponsored by Claire Daniels at Walter Reed National Military Medical Center to determine if dual therapy is superior to fluticasone monotherapy in children and adults.
