Fluzoparib (also known as fuzuloparib) is an orally bioavailable small molecule inhibitor of poly(ADP-ribose) polymerase types 1 and 2 (PARP1 and PARP2). It acts by inhibiting PARP-mediated repair of damaged DNA via the base excision repair pathway, leading to accumulation of DNA strand breaks, genomic instability, and induction of apoptosis in tumor cells. This mechanism enhances the cytotoxicity of DNA-damaging agents and may reverse tumor cell chemoresistance and radioresistance. Fluzoparib is primarily developed for treatment of solid tumors including ovarian cancer, fallopian tube cancer, primary peritoneal cancer, breast cancer, pancreatic cancer, prostate cancer, glioblastoma, and small cell lung cancer. It has been approved in China for platinum-sensitive recurrent ovarian cancer with germline BRCA mutations after second-line or later chemotherapy.
