Fluzoparib + camrelizumab is an investigational combination therapy consisting of two agents: - Fluzoparib is a potent, orally available small molecule inhibitor of poly(ADP-ribose) polymerase (PARP), primarily PARP1. It induces synthetic lethality in tumor cells with homologous recombination repair (HRR) deficiencies such as BRCA mutations by inhibiting DNA repair and promoting cell death. Fluzoparib has demonstrated antitumor activity in preclinical models and clinical trials for various solid tumors[6][8][10]. - Camrelizumab is a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance T-cell mediated antitumor immunity. The combination aims to exploit the synergistic effects of DNA damage induction by fluzoparib and immune activation by camrelizumab. This regimen is under investigation for several advanced cancers including recurrent/metastatic nasopharyngeal carcinoma, TP53-mutated endometrial cancer, and advanced melanoma with HR pathway gene alterations[2][4][5][7].
