This is a combination therapy consisting of three agents: - **Fluzoparib** is an orally available small-molecule inhibitor of poly(ADP-ribose) polymerase (PARP), primarily PARP1, which impairs DNA repair in cancer cells, especially those with homologous recombination deficiency. It induces DNA double-strand breaks and apoptosis in HR-deficient cells and sensitizes both HR-deficient and proficient cells to cytotoxic drugs[1][2][5]. - **Camrelizumab** is a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance T-cell mediated anti-tumor immunity. - **Temozolomide** is an oral alkylating agent that damages tumor cell DNA by methylation, leading to inhibition of replication and cell death. It is widely used for the treatment of brain tumors such as glioblastoma[6][9]. The combination aims to exploit synthetic lethality (via PARP inhibition), immune activation (via PD-1 blockade), and direct cytotoxicity (via alkylation) for synergistic anti-cancer effects. This regimen has been studied in advanced/metastatic melanoma with homologous recombination pathway gene mutations/alterations[10].
