fluzoparib + nab-paclitaxel is a combination therapy comprising fluzoparib, a potent oral poly (ADP-ribose) polymerase (PARP) inhibitor, and nab-paclitaxel, a nanoparticle albumin-bound formulation of the microtubule-stabilizing agent paclitaxel. Fluzoparib selectively inhibits PARP1 and PARP2, enzymes critical for the repair of single-strand DNA breaks via the base excision repair pathway. In cells with homologous recombination repair (HRR) deficiencies, such as those with BRCA mutations, PARP inhibition leads to the accumulation of double-strand breaks and subsequent cell death (synthetic lethality). Nab-paclitaxel enhances the delivery of paclitaxel to tumor tissues, where it stabilizes microtubules and inhibits mitotic progression. The combination is hypothesized to exert synergistic anti-tumor effects by increasing DNA damage and disrupting the cell cycle. This regimen is currently being evaluated in Phase II clinical trials for the treatment of metastatic castration-resistant prostate cancer (mCRPC), with research led by institutions including Shandong Cancer Hospital and Institute.
