Overview

FMC63-KIR-CAR is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target the CD19 antigen, which is widely expressed in B-cell malignancies. It utilizes the well-characterized FMC63 single-chain variable fragment (scFv) for antigen recognition, integrated into a multi-chain natural killer immunoglobulin-like receptor (KIR)-based signaling platform. Developed by Verismo Therapeutics, the KIR-CAR platform mimics the physiological signaling of natural killer (NK) cells by separating antigen binding from activation signals, a design intended to reduce the constitutive T-cell activation and exhaustion often observed with conventional CD3-zeta-based CAR-T therapies. Preclinical studies have demonstrated that FMC63-KIR-CAR achieves faster and deeper anti-tumor responses with significantly lower systemic cytokine production compared to traditional 4-1BB/CD3-zeta CAR-T cells, suggesting a potentially improved safety and efficacy profile for patients with relapsed or refractory B-cell non-Hodgkin lymphoma (B-NHL).
