FMNL1-overexpressing CAR-T cells are an engineered adoptive cellular therapy designed to overcome the physical barriers of the solid tumor microenvironment. By overexpressing Formin-like-1 (FMNL1), a key regulator of the actin cytoskeleton and T cell migration, these cells exhibit enhanced motility and infiltration capabilities. This modification allows the T cells to better penetrate dense extracellular matrix and abnormal vasculature, which typically limit the efficacy of standard CAR-T therapies in solid tumors. Preclinical data presented at AACR 2026 demonstrated that these cells significantly increase accumulation at tumor sites and prolong survival in murine models of melanoma and lung carcinoma, regardless of the specific CAR construct or tumor antigen used.
