FMP014 is a self-assembling protein nanoparticle (SAPN) vaccine candidate developed for the prevention of malaria caused by Plasmodium falciparum. The vaccine consists of 60 identical monomer protein chains that form an icosahedral nanoparticle structure. Each monomer incorporates selected CD4+ and CD8+ T cell epitopes from the P. falciparum circumsporozoite protein (PfCSP), universal T helper epitopes, portions of the α-thrombospondin repeat (α-TSR) domain, and six repeats of the NANP motif from PfCSP. When formulated with Army Liposomal Formulation containing QS21 saponin adjuvant (ALFQ), FMP014 elicits strong antibody responses targeting both NANP repeat and C-terminal peptides, inhibits sporozoite invasion in vitro, and demonstrates protective efficacy in animal models. The vaccine has undergone Phase 1 clinical evaluation for safety, immunogenicity, tolerability, and preliminary efficacy in healthy adults[1][2][3][4][6].
