FMP374 is a novel, orally bioavailable small molecule dual-acting N-methyl-D-aspartate receptor (NMDAR) modulator being developed by FundaMental Pharma GmbH. It is specifically designed for the treatment of treatment-resistant depression (TRD), a condition affecting a significant portion of patients with major depressive disorder who do not respond to standard therapies. Unlike traditional NMDAR antagonists such as ketamine, FMP374 has demonstrated rapid and sustained antidepressant effects in preclinical models at low nanomolar concentrations without inducing dissociation-like behaviors, hallucinations, ataxia, or hyperactivity at therapeutic doses. This favorable safety profile positions FMP374 as a potential at-home oral therapy.
