Overview

FN14 siRNA (also known as siFn14) is a small interfering RNA designed to silence the expression of the fibroblast growth factor-inducible 14 (Fn14) receptor, also known as TNFRSF12A or the TWEAK receptor. Fn14 is a member of the tumor necrosis factor receptor superfamily and is the primary receptor for the cytokine TWEAK (TNF-like weak inducer of apoptosis). The TWEAK/Fn14 signaling pathway plays a critical role in regulating inflammation, cell proliferation, and tissue remodeling. Overexpression of Fn14 is associated with various inflammatory and fibrotic diseases, including psoriasis, rheumatoid arthritis, and cutaneous lupus erythematosus. By utilizing RNA interference to knockdown Fn14, this therapeutic approach aims to suppress downstream proinflammatory signaling (such as the NF-κB and MAPK pathways) and reduce the production of inflammatory cytokines and chemokines. Preclinical studies have investigated various delivery methods, including transdermal composite ionic liquids and lentiviral vectors, for indications such as skin lesions, spinal cord injury, and vascular dysfunction.
