Folate binding protein E39 is an investigational cancer vaccine composed of the HLA-A2–restricted peptide E39 (FBP amino acids 191–199, sequence EIWTHSYKV) derived from folate binding protein (folate receptor alpha), typically administered with the immune adjuvant granulocyte macrophage–colony stimulating factor (GM‑CSF) to induce FBP‑specific cytotoxic T‑lymphocyte responses in patients with FBP‑overexpressing malignancies such as ovarian, endometrial, and breast cancer. Clinical phase I/IIa and Ib trials in disease‑free high‑risk ovarian, endometrial, and breast/ovarian cancer patients have shown that E39+GM‑CSF is safe, elicits dose‑dependent in vivo immune responses (e.g., delayed‑type hypersensitivity and expansion of E39‑specific CTLs), and may reduce recurrence risk, particularly at a 1000 mcg peptide dose, supporting further development as an adjuvant immunotherapy. The program has been developed primarily by Galena Biopharma in collaboration with academic centers, including the University of Texas MD Anderson Cancer Center.
