Folate-conjugated immunoglobulin is a chemically modified antibody in which folic acid (folate) molecules are covalently attached to immunoglobulin (IgG), usually via amide bonds. This enables the antibody to specifically target cells expressing high levels of the folate receptor (FR), which is overexpressed in many tumor types—including ovarian, breast, lung, endometrial, and colon cancers. The folate ligand facilitates receptor-mediated endocytosis, enhancing selectivity for FR-positive cells. The IgG backbone enables effector functions such as antibody-dependent cellular cytotoxicity (ADCC) by natural killer (NK) cells, leading to tumor cell lysis. The conjugate acts as a targeted immunotherapy for FR-positive malignancies and has demonstrated increased antitumor efficacy, especially when combined with immunostimulatory agents. The construct may be composed of human or murine IgG, typically carrying 2–3 folate moieties per IgG molecule for optimal activity and reduced effector impairment. This agent is largely experimental and not an approved product, mainly used in preclinical and early-stage cancer research[1][3][5].
