Drug intelligence / Profile preview
Folate-targeted liposomal methotrexate is an experimental nanomedicine developed by Solfarcos for the treatment of rheumatoid arthritis (RA). The formulation consists of methotrexate encapsulated within liposomes that are surface-functionalized with folate ligands. This design exploits the overexpression of folate receptor beta (FR-β) on activated macrophages, which are key drivers of synovial inflammation in RA. By selectively targeting these cells, the drug aims to enhance therapeutic efficacy within the joints while minimizing the systemic adverse effects typically associated with conventional methotrexate therapy, such as gastrointestinal toxicity and hepatotoxicity. Once internalized by the target cells, methotrexate acts as a dihydrofolate reductase (DHFR) inhibitor, disrupting folate metabolism and suppressing inflammatory responses.
