FOLFOX7 is a specific regimen of the combination chemotherapy known as FOLFOX, primarily used for the treatment of metastatic colorectal cancer. The regimen consists of three drugs: oxaliplatin (a platinum-based chemotherapeutic that forms DNA crosslinks and inhibits DNA synthesis), leucovorin calcium (folinic acid, which enhances the effectiveness of fluorouracil), and fluorouracil (5-FU, an antimetabolite that interferes with DNA and RNA synthesis). In the FOLFOX7 protocol, higher doses of oxaliplatin are administered compared to some other variants. This combination targets rapidly dividing cancer cells through multiple mechanisms—oxaliplatin causes DNA damage via crosslinking; 5-FU disrupts both RNA function and DNA synthesis by incorporating into nucleic acids or inhibiting thymidylate synthase; leucovorin potentiates 5-FU’s effects. The regimen is typically given in cycles every two weeks intravenously. It is especially used as first-line therapy for patients with potentially resectable or unresectable metastatic colorectal cancer[1][6][8].
