Overview
FOLFOXIRI + panitumumab is a combination regimen used primarily for the first-line treatment of metastatic colorectal cancer (mCRC) in patients with wild-type RAS and BRAF genes. FOLFOXIRI is a chemotherapy regimen that includes 5-fluorouracil, leucovorin, oxaliplatin, and irinotecan. Panitumumab is a fully human monoclonal antibody targeting the epidermal growth factor receptor (EGFR). Panitumumab binds specifically to EGFR on both normal and tumor cells and competitively inhibits ligand binding. This prevents receptor autophosphorylation and downstream signaling involved in cell proliferation and survival. The addition of panitumumab to FOLFOXIRI aims to enhance antitumor activity by combining cytotoxic chemotherapy with targeted inhibition of EGFR-mediated pathways[1][2][5][6]. The combination has been studied in phase III clinical trials for unresectable metastatic colorectal cancer[5].
