Overview
Foliglurax is a small molecule drug that acts as a positive allosteric modulator of the metabotropic glutamate receptor 4 (mGluR4). It was developed for the treatment of Parkinson's disease and drug-induced dyskinesia. The mechanism of action involves enhancing mGluR4 activity to normalize inhibitory and excitatory signaling in basal ganglia pathways, potentially reducing motor symptoms and levodopa-induced dyskinesia. Foliglurax advanced to phase II clinical trials but failed to meet primary efficacy endpoints compared to placebo. Development was discontinued after these results[1][5][6].
