Overview
This is a four-drug combination regimen consisting of folinic acid (leucovorin), fluorouracil (5-FU), oxaliplatin, and aflibercept. Folinic acid, fluorouracil, and oxaliplatin together are commonly known as FOLFOX, a standard chemotherapy protocol for colorectal cancer. Aflibercept is a recombinant fusion protein that acts as a decoy receptor for vascular endothelial growth factor (VEGF), inhibiting angiogenesis. The addition of aflibercept to the FOLFOX regimen aims to enhance anti-tumor efficacy by combining cytotoxic chemotherapy with targeted anti-angiogenic therapy. This combination has been investigated in metastatic colorectal cancer (mCRC) settings, particularly after progression on prior therapies[1]. The mechanism involves direct cytotoxicity from the chemotherapeutic agents and inhibition of tumor blood vessel formation by aflibercept.
